The effects of taurine on glutamate metabolism in rats after acute manganese exposure

摘要: 目的: 探讨牛磺酸对急性染锰大鼠脑内谷氨酸代谢的影响。方法: 选取清洁级Wistar大鼠45只,随机分为对照组、模型组和治疗组,每组各15只。对照组腹腔注射剂量为30 mg/kg体质量的生理盐水,1次/d,共20次。模型组腹腔注射剂量为15 mg/kg体质量的MnCl2·4H2O溶液,1次/d,共20次。治疗组在模型组基础上,皮下注射剂量为125.15 mg/kg体质量的牛磺酸,1次/隔日,共10次。进行Morris水迷宫定位航行实验和跳台实验,检测丘脑中谷氨酰胺酶(PAG)、谷氨酸脱羧酶(GAD)、谷氨酰胺合成酶(GS)含量和脑组织中超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果: 与对照组比较,模型组定位航行实验的潜伏期更长、错误次数更多(均P<0.05),跳台实验4 d的潜伏期均明显延长(均P<0.05),丘脑中PAG、GAD、GS水平均明显降低(均P<0.01),脑组织中SOD明显降低、MDA明显增高(均P<0.05)。与模型组比较,治疗组定位航行实验的潜伏期明显缩短、错误次数明显减少(均P<0.05),跳台实验4 d的潜伏期均明显缩短(均P<0.05),丘脑中PAG、GAD、GS水平均明显增高(均P<0.01),脑组织中SOD明显增高、MDA明显降低(均P<0.05)。结论: 牛磺酸对急性染锰大鼠脑组织具有保护作用,能调节PAG、GAD和GS的活性,抑制脑组织过氧化反应,提高学习记忆能力。Abstract: Objective: The effects of taurine on glutamate metabolism in the brains of rats after acute manganese exposure were studied.Method: Forty-five Wistar rats were randomly divided into control group,model group and treatment group,with 15 rats in each group.In control group,the rats were receiving intraperitoneal injection of physiological saline at the dose of 30 mg/kg bw once a day,for 20 times.In model group,the rats were receiving intraperitoneal injection of MnCl2·4H2O at the dose of 15 mg/kg bw once a day,for 20 times.In treatment group,on the basis of model group,the rats were receiving subcutaneous injection of taurine at the dose of 125.15 mg/kg bw once a day,for 10 times.Morris water navigation task and step-down test were done in three groups.The levels of Phosphate activated glutaminase(PAG),glutamic acid decarboxylase(GAD),glutamine synthetase(GS)in thalamus,and super oxide dismutase(SOD),malondialdehyde(MDA)in brain tissue were measured.Result: Comparing with control group,model group had longer incubation period and more wrong search results in Morris water navigation task,and longer incubation periods in step-down test for 4 days(all P<0.05).PAG,GAD,GS(all P<0.01) and SOD(P<0.05) of model group were significantly lower than those of control group,while MDA was higher than that of control group(P<0.05).Comparing with model group,treatment group had shorter incubation period and lower error frequency in Morris water navigation task,and shorter incubation period in step-down test for 4 days(all P<0.05).PAG,GAD,GS(all P<0.01) and SOD(P<0.05) of treatment group were significantly higher than those of model group,while MDA was lower than that of model group(P<0.05).Conclusion: Taurine have the potential to regulate the activity of PAG,GAD and GS in rats with acute manganese exposure,and to inhibit peroxidation of brain tissue and improve the ability of learning and memory.
