Relationship between the expression levels of plasma long noncoding RNA THRIL and NEAT1 and sepsis complicated with acute respiratory distress syndrome

摘要: 目的:探讨血浆长链非编码RNA-肿瘤坏死因子相关异种核核糖核蛋白L(lncRNA THRIL)、核富集转录本1(lncRNA NEAT1)表达水平与脓毒症并发急性呼吸窘迫综合征(ARDS)的关系。方法:纳入我院2017年5月—2020年5月期间收治的脓毒症患者110例,根据其是否合并ARDS分成ARDS组(n=42)、非ARDS组(n=68)。比较两组血浆lncRNA THRIL、lncRNA NEAT1表达水平,经logistic多元回归分析患者发生ARDS的危险因素。根据脓毒症并发ARDS患者住院期间的预后分成生存组、死亡组,比较两组血浆lncRNA THRIL、lncRNA NEAT1表达水平。经Pearson线性相关分析二者的相关性,绘制受试者工作特征曲线(ROC)分析其评价患者预后的曲线下面积(AUC)。结果:ARDS组血浆lncRNA THRIL、lncRNA NEAT1表达水平高于非ARDS组,生存组血浆lncRNA THRIL、lncRNA NEAT1水平低于死亡组(P<0.05)。Logistic多元回归分析提示,重症胰腺炎、较高的血浆lncRNA THRIL和lncRNA NEAT1水平均是脓毒症患者ARDS发生的危险影响因素(P<0.05)。血浆lncRNA THRIL与lncRNA NEAT1呈正相关(P<0.05)。ROC分析提示,血浆lncRNA THRIL、lncRNA NEAT1对脓毒症并发ARDS预后有较高的评估价值,AUC(95%CI)分别为0.741(0.611~0.899)、0.711(0.522~0.968)。联合应用价值更高,AUC(95%CI)为0.852(0.750~0.968)。结论:脓毒症并发ARDS患者的血浆lncRNA THRIL、lncRNA NEAT1表达水平明显增高,二者有望成为评估患者预后的辅助指标。

Abstract: Objective: To investigate the relationship between the expression levels of plasma lncRNA-Tumor necrosis factor related heterogeneous nuclear ribonucleoprotein L-related immunoregulatory(lncRNA THRIL), long noncoding RNA nuclear-enriched abundant transcript 1(lncRNA NEAT1) and sepsis complicated with acute respiratory distress syndrome(ARDS).Methods: Include 110 sepsis patients who were admitted to our hospital from May 2017 to May 2020. According to complicated by combined with ARDS or not, they were divided into ARDS group(n=42) and non-ARDS group(n=68). Expression levels of plasma lncRNA THRIL, lncRNA NEAT1 in the two groups were compared, and the risk factors of ARDS were analyzed by logistic multiple regression analysis. According to the prognosis of patients with sepsis complicated by ARDS during hospitalization, they were divided into survival group and death group. The plasma lncRNA THRIL and lncRNA NEAT1 expression levels in the two groups were compared, Pearson linear correlation was used to analyze their correlation, The subjects' working characteristic curve(ROC) was drawn to analyze the area under curve(AUC) of the patients' prognosis.Results: The expression levels of plasma lncRNA THRIL and lncRNA NEAT1 in the ARDS group were higher than those in the non-ARDS group, and levels of plasma lncRNA THRIL and lncRNA NEAT1 in the survival group were lower than those in the death group(P<0.05). Logistic multiple regression analysis indicated that severe pancreatitis, higher levels of plasma lncRNA THRILandlncRNA NEAT1 were risk factors for ARDS in sepsis patients(P<0.05). There was a positive correlation between plasma lncRNA THL and lncRNANEAT1(P<0.05). ROC analysis indicated that plasma lncRNA THRIL and lncRNA NEAT1 had a high evaluation value for the prognosis of sepsis complicated by ARDS, with AUC(95%CI) of 0.741(0.611-0.899) and 0.711(0.522-0.968), respectively. The combined application value was even higher, with AUC(95%CI) of 0.852(0.750-0.968).Conclusion: The expression levels of plasma lncRNA THRIL and lncRNA NEAT1 in patients with sepsis complicated with ARDS are significantly increased, which are promising to be an auxiliary indicator for evaluating the prognosis of patients. -
