Correlation study on expression of microRNA-21 in peripheral blood and prognosis of paraquat poisoning

摘要: 目的: 探讨外周血microRNA的表达变化对百草枯中毒患者预后的相关性。方法: 选择2016-07-2017-12期间我科收治的57例百草枯中毒患者,依据28 d是否存活分为生存组和死亡组。RT-PCR检测生存组和死亡组患者入科24 h血清中miR-21、miR-29、miR-155、Let-7 d的水平;ELISA法检测两组患者入科24 h血清中miRNA相关细胞因子,分析临床指标与百草枯中毒预后的相关性。结果: 生存组25例,死亡组32例。与生存组相比,死亡组患者入科24 h外周血中miRNA-21、miR-29、miR-155表达显著升高,Let-7 d则明显下降;死亡组患者血清中IL-10显著下降,TGF-β则无明显变化。死亡组24 h内的中毒剂量,尿素氮(BUN),谷丙转氨酶(ALT),白细胞计数(WBC),动脉血二氧化碳分压(PaCO2),血乳酸(Lac),miR-21,IL-10等指标明显高于生存组,miR-21水平与患者生存时间负相关。结论: 外周血中miRNA-21可作为早期预测百草枯中毒患者预后的指标。

Abstract: Objective: To investigate the value of peripheral blood microRNA in predicting the severity and prognosis of pulmonary fibrosis in patients with paraquat poisoning. Method: Select 57 cases of paraquat poisoning patients from July 2016 to December 2017 admitted to our department.these patients were divided into survival group and death group according to their outcome.RT-PCR detection of miR-21,miR-29,miR-155,Let-7 d expressions in serum of patients with survivors and death patients in 24 hours after admission,ELISA detection of miRNA-related cytokines in the serum of two groups in 24 hours after admission. Result: Twenty-five patients in survival group,32 cases in death group.Compared with the survival group,in 24 hours after admission,the expression of miRNA-21,miR-29,and miR-155 in the peripheral blood of the death group was significantly higher than that in the survival group,while the expression of Let-7 d decreased significantly.IL-10 in the serum of the death group decreased significantly,while TGF-beta did not change significantly.In death group BUN,ALT,WBC,PaCO2,Lac,miR-21,IL-10 were significantly higher than those in the survival group,and the miR-21 level was negatively correlated with the survival time of the patients.Conclusion: miRNA-21 in peripheral blood can be used as an early predictor of prognosis in patients with paraquat poisoning.-
